Is it possible to use a voltage divider on EN pin so output is enabled when VIN = 4.5V and disabled when VIN = 2V? Should I use the EN input current of 2.1µA to set hysteresis?

Yes, you can use voltage divider from Vin to EN. Typically, the input current to EN pin is 2.1uA. So, yes using 2.1uA input current should be okay.
